In vitro regeneration of bulblet using two and four bulb-scales explants of summer snowflake (Leucojum aestivum L.)

Abstract Leucojum aestivum is a valuable and endangered plant species with bulb scales best suited as explants in micropropagation. In the current study, its micropropagation was investigated by using two different various concentrations combinations of growth regulators (PGRs). Bulbs were first disinfected benomyl® for 5 hours. After meeting chilling requirements, two-scale four-scale provided direct indirect organogenesis. Explants exposed to hot water, 70% ethanol 2.5% sodium hypochlorite further disinfestation. Four-scale treated naphthaleneacetic acid (NAA), 6-benzyladenine (BA), kinetin (Kin) bulblet regeneration. For callogenesis, 0.5 mg L-1 BA combined 1, 2, 3, 4, or 6 2,4-dichlorophenoxyacetic (2,4-D) applied. Regarding explants, BA, Indole-3-butyric (IBA) NAA used induction, Indoleacetic (IAA), NAA, 4-D callus induction. None responded regeneration induction media. Unlike, regenerated bulblets roots control medium MS media enriched PGRs. Callus generated on supplemented 2,4-D observed some cases. On medium, had natural form, but PGRs-rich media, they deformed. percentage, number length root length, no significant differences found between PGR-treatment each case. Therefore, it seems logical suggesting not use PGRs, which will considerably reduce costs at large-scale production.
